More Than a Decade of Research on Clonal Hematopoiesis and Cardiovascular Disease

Men typically have XY sex chromosomes, but with aging, the Y chromosome can sometimes be lost from blood leukocytes (although an individual’s gender identity does not always correspond to their chromosomal composition). This phenomenon, known as loss of Y chromosome (LOY), has been associated with age-related diseases such as Alzheimer’s disease and cancer. Dr. Soichi Sano conducts research on the relationship between LOY and heart failure, a condition in which the heart’s function is impaired.

Lecture Overview

It is now well recognized that our cells gradually accumulate genetic mutations with age, and some of these mutations expand to form “clones.” When this occurs in blood cells, it is referred to as clonal hematopoiesis (CH). Recent studies have revealed that CH increases the risk of various age-related diseases, including cardiovascular conditions such as heart failure and atherosclerosis, attracting significant global attention.

The first report linking CH to cardiovascular disease was published in 2014 from Boston, where I was conducting research at the time. Over the past decade, this field has rapidly advanced—from basic science to clinical epidemiology and even toward potential therapeutic applications. In this lecture, I will provide a clear overview of the research developments over the past ten years and discuss future perspectives in this emerging field.
